ERL, who built my block, said to turn it on, make sure you have oil pressure, let it warm up to operating temp., then drain the oil, change the filter, then tune immediately. However, I never got a straight answer on whether or not to break it in/tune it w/ boost. Most of the people that I ask say to break it in with boost, just wanted some more opinions. I believe one of the most important matters during break-in besides watching vitals (AFR, Oil Press, Temp...) is to make sure that the engine is feeling load, such as a good range of varying accelleration/rpm while the car is in motion, to get the rings to seat against the cylinder walls.
